Standard Operating Procedure (SOP): Operation of Polarimeter

Purpose:
The purpose of this Standard Operating Procedure (SOP) is to provide detailed instructions for the proper operation of a Polarimeter. This SOP ensures accurate and reliable measurements of optical rotation or specific rotation of optically active substances in solution. Polarimetry is commonly used in pharmaceutical, chemical, and food industries for quality control, identification, and characterization of chiral compounds.

Scope:
This SOP applies to all personnel involved in operating and maintaining the Polarimeter in a laboratory or testing environment.

Responsibility:
Laboratory personnel: Responsible for performing polarimetry measurements as per this SOP.
Quality Assurance (QA): Responsible for reviewing and approving this SOP.

Procedure:
4.1. Equipment and Materials:
Polarimeter
Sample solutions of known concentration and optical rotation
Sample cells or tubes suitable for polarimetry measurements
Suitable cleaning solutions or solvents
Relevant safety equipment (e.g., gloves, safety glasses)
4.2. Pre-Operational Preparations:
4.2.1. Ensure that the Polarimeter is clean, free from dust or residues, and properly maintained.
4.2.2. Familiarize yourself with the specific operating instructions and technical specifications provided by the manufacturer for the particular model of the Polarimeter being used.
4.2.3. Check the availability and condition of the required sample solutions of known optical rotation.
4.2.4. Gather all necessary equipment

and materials.
4.3. Instrument Calibration:
4.3.1. Perform the necessary calibration procedures as specified by the manufacturer or the laboratory’s standard operating procedures. This involves calibrating the Polarimeter using standard solutions of known optical rotation and adjusting the instrument accordingly.
4.3.2. Document the calibration process and results as per the laboratory’s document control procedures.
4.4. Sample Measurement Procedure:
4.4.1. Prepare the sample solutions, ensuring they are in a suitable form and concentration for polarimetry measurements.
4.4.2. Set up the Polarimeter according to the manufacturer’s instructions, including the proper selection of wavelength, temperature control (if applicable), and sample cell or tube suitable for the specific measurement.
4.4.3. Ensure that the instrument is warmed up and stabilized before starting the measurement.
4.4.4. Rinse the sample cell or tube with suitable cleaning solutions or solvents and carefully blot it dry before each measurement.
4.4.5. Fill the sample cell or tube with the prepared sample solution, ensuring there are no air bubbles or impurities that may affect the measurement.
4.4.6. Place the sample cell or tube into the Polarimeter and record the initial reading displayed.
4.4.7. Rotate the polarizer or adjust the instrument controls to achieve a dark background, indicating the absence of optical rotation.
4.4.8. Slowly rotate the analyzer or adjust the instrument controls to observe the rotation of the polarized light by the sample.
4.4.9. Record the final reading displayed, indicating the observed optical rotation.
4.4.10. If multiple samples are to be analyzed, rinse the sample cell or tube with suitable cleaning solutions or solvents between measurements to prevent cross-contamination.
4.5. Post-Measurement Procedures:
4.5.1. Rinse the sample cell or tube with suitable cleaning solutions or solvents and carefully blot it dry after each measurement to remove any residue.
4.5.2. Clean the sample cell or tube using appropriate cleaning procedures recommended by the manufacturer or the laboratory’s standard operating procedures.
4.5.3. Store the Polarimeter and sample cells or tubes in a clean and secure location following the manufacturer’s guidelines for proper storage.

Records:
5.1. Maintain a record of the calibration process, including the date, standard solutions used, instrument adjustments, and calibration curve (if applicable).
5.2. Document the details of each polarimetry measurement conducted, including the date, sample identification, wavelength, temperature (if applicable), initial and final readings, and any relevant remarks or observations.
5.3. Retain the records in a secure and organized manner as per the laboratory’s document control procedures.
