2. Scope
This SOP applies to the Analytical Method Development (AMD) department and is intended for solid APIs, excipients, and processed materials requiring surface area characterization during R&D, quality control, and stability testing phases.
3. Responsibilities
- Analytical Scientist: Develops and validates the BET method, selects appropriate degassing and analysis parameters.
- Instrument Analyst: Prepares samples, operates the surface area analyzer, and records data.
- QA Officer: Ensures reviewed data complies with regulatory standards.
- Head – AMD: Authorizes the validated method and ensures integration into product development workflows.
4. Accountability
The Head of AMD is accountable for ensuring the BET method is scientifically robust, reproducible, and compliant with ICH and USP requirements.
5. Procedure
5.1 Instrument Calibration
- Use a certified reference material (e.g., alumina or silica with known surface area).
- Calibrate the instrument for:
- Dead volume
- Leak check
- Flow rate accuracy
- Record all calibration data in Annexure-1: BET Calibration Log.
5.2 Sample Preparation
- Weigh 0.2–1.0 g of accurately measured dry sample into the analysis tube.
- Degas sample under vacuum or flowing nitrogen at suitable temperature (e.g., 100°C–150°C) for 2–6 hours depending on sample sensitivity.
- Document degassing conditions in Annexure-2: Sample Prep Log.
5.3 Method Parameters
- Adsorbate Gas: Nitrogen (N₂)
- Temperature: Liquid nitrogen bath (~77 K)
- Relative Pressure (P/P₀): 0.05–0.3 for BET range
- BET Equation: Used to calculate monolayer capacity (Vm) and surface area (m²/g)
- Number of Data Points: Minimum 5–7 in linear BET region
5.4 Analysis and Calculation
- Initiate adsorption analysis and collect isotherm data (adsorption and desorption branches).
- Plot BET graph and ensure linearity in selected P/P₀ range (R² ≥ 0.995).
- Calculate:
- Monolayer capacity (Vm)
- Specific surface area using molecular cross-sectional area of nitrogen (0.162 nm²)
- Log surface area results in Annexure-3: Surface Area Report.
5.5 Method Validation
- Validate according to USP <846> and ICH Q2(R2) guidance:
- Repeatability: Surface area results within ±3% across 3 replicate measurements
- Specificity: No interference from impurities or moisture
- Linearity: Ensure BET region fits R² ≥ 0.995
- Robustness: Assess effect of degassing time and sample mass
- Summarize validation in Annexure-4: BET Validation Summary.
5.6 Critical Points and Troubleshooting
- Ensure proper degassing to avoid low surface area readings.
- Avoid moisture exposure post-degassing.
- Ensure liquid nitrogen levels remain stable during analysis.
